{"id":57809,"date":"2017-01-25T08:54:02","date_gmt":"2017-01-25T07:54:02","guid":{"rendered":"https:\/\/www.irta.cat\/noticia\/a-new-form-of-prion-replication-is-described-that-might-explain-some-transmission-barriers\/"},"modified":"2024-11-06T09:19:23","modified_gmt":"2024-11-06T08:19:23","slug":"a-new-form-of-prion-replication-is-described-that-might-explain-some-transmission-barriers","status":"publish","type":"noticia","link":"https:\/\/www.irta.cat\/en\/noticia\/a-new-form-of-prion-replication-is-described-that-might-explain-some-transmission-barriers\/","title":{"rendered":"A new form of prion replication is described that might explain some transmission barriers"},"content":{"rendered":"<p><b><span lang=\"EN-US\">Part of the <i>in vivo<\/i> experiments of this international study were performed in IRTA-CReSA level 3 biocontainment facility <\/span><\/b><\/p>\n<p><span lang=\"EN-US\">A new form of prion replication is described for the first time in a study recently published in the journal Proceedings of the National Academy of Sciences (PNAS) in which prions are able to propagate in the new host but are not adapted and preserve Its original characteristics:\u00a0<\/span><b><span lang=\"EN-US\">NAPA or non-adaptive prion amplification<\/span><\/b><span lang=\"EN-US\">.<\/span><\/p>\n<p><span lang=\"EN-US\">A considerable\u00a0part of the\u00a0<\/span><i><span lang=\"EN-US\">in vivo<\/span><\/i><span lang=\"EN-US\">\u00a0experiments with mice of this study have been carried out in IRTA-CReSA\u2019s \u00a0level 3 biocontainment unit and in PRIOCAT laboratory, both \u00a0meet the biosecurity conditions necessary to work with these infectious agents. This unit of biocontention is part of the <a href=\"http:\/\/www.rlasb.es\/\">RELASB<\/a>, the network of high biological safety laboratories in the map of\u00a0<a href=\"http:\/\/www.idi.mineco.gob.es\/portal\/site\/MICINN\/menuitem.eed4570ef37d2c8fbaa777b9026041a0\/?vgnextoid=b8be18ef1a553510VgnVCM1000001d04140aRCRD&amp;lang_choosen=es\">Unique Scientific and Technical Infrastructures (ICTS)<\/a>\u00a0in Spain. <\/span><\/p>\n<p><span lang=\"EN-US\">The in vitro experiments were carried out by the group of Dr. Joaquin Castilla of the CICbioGUNE in Bilbao. The study was led by the group of Dr. Glenn Telling of the Colorado Prion Research Center (PRC) in the United States and included the collaboration of other US and Italian groups.<\/span><\/p>\n<p><span lang=\"EN-US\">The study describes, among other examples, how scrapie prions (a TSE affecting sheep) can be replicated inefficiently in transgenic mice that have the prion protein of the horse and yet are unable to adapt and continue to propagate in this species. Instead, they can be replicated efficiently in a mouse that expresses the sheep prion protein. That is, they retain the properties of the original inoculum. <\/span><\/p>\n<p><span lang=\"EN-US\">The study has also shown, for the first time, that\u00a0<\/span><b><span lang=\"EN-US\">horses are resistant to infection by several different strains of prions.<\/span><\/b><span lang=\"EN-US\">\u00a0It seems that this is due to a special conformation of its prion protein.<\/span><\/p>\n<div id=\"attachment_10475\" style=\"width: 730px\" class=\"wp-caption aligncenter\"><img loading=\"lazy\" decoding=\"async\" aria-describedby=\"caption-attachment-10475\" class=\"wp-image-10475 size-full\" src=\"https:\/\/www.irta.cat\/wp-content\/uploads\/2023\/01\/figura-prions-eng-1.jpg\" alt=\"\" width=\"720\" height=\"405\" \/><p id=\"caption-attachment-10475\" class=\"wp-caption-text\">Autor: Enric Vidal Figure: Some of the experiments performed with transgenic mice expressing the horse prion protein. Red: efficient transmission<\/p><\/div>\n<p><span lang=\"EN-US\">This new model of prion replication has also been observed in Transmissible Mink Encephalopathy (TME) transmission experiments in transgenic mice expressing the deer prion protein.<\/span><\/p>\n<p><span lang=\"EN-US\">Interestingly, this model could explain the inefficient transmission of BSE, Bovine Spongiform Encephalopathy or mad cow disease, to humans \u2026 the hypothesis is that this was another case of NAPA. Humans can\u00a0be infected with BSE but this prion does not adapt to\u00a0our species.
